Skeletal muscle tissue regulates locomotion, metabolic homeostasis, and systemic glucose utilization and is highly sensitive to inflammatory, hypoxic, and metabolic stress. Cross-talk between myocytes, stromal fibroblasts, endothelial cells, adipocytes, and immune populations strongly affects tissue regeneration, extracellular matrix remodeling, cytokine secretion, and progression of muscle injury. Disturbance of this microenvironment may lead to chronic inflammation, fibrosis, impaired contractility, and metabolic dysfunction. Therefore, incorporation of multiple native cellular compartments is highly essential for predictive modelling of muscle toxicity and disease progression.
